Abstract

The present invention provides a purified and isolated DNA-binding protein, HPBF, which specifically binds to the promoter region of the Her-2/neu (ERBB2/c-erbB-2) gene sequence, the presence of which provides an early indication of transition to a cancerous state has been found. The present invention also provides bioassays for screening substances for the ability to inhibit HPBF activity, the ability to inhibit the mitogenic activity of HPBF and the ability to inhibit HPBF production. The present invention further provides methods of inhibiting the biological activity mediated by HPBF comprising preventing the HPBF from binding to the promoter region of the ERBB2 gene sequence.
La présente invention concerne une protéine de liaison d'ADN purifiée et isolée, dénommée HPBF, qui se fixe spécifiquement au promoteur de la séquence du gène Her-2/neu (ERBB2/c-erbB-2), dont la présence donne une indication précoce du passage à un état cancéreux. La présente invention concerne également des biodosages permettant de trier des substances en fonction de leur aptitude à inhiber l'activité de la protéine HPBF, de leur aptitude à inhiber l'activité mytogénique de la protéine HPBF, et de leur aptitude à inhiber la production de protéine HPBF. La présente invention concerne également des procédés permettant d'inhiber l'activité biologique due à la présence de la protéine HPBF, consistant à empêcher la protéine HPBF de se fixer au promoteur de la séquence du gène ERBB2.
